A pele é dotada de caminhos metabólicos e receptores inatos para protegê-la contra ataques externos. Neste artigo, descrevemos um componente protetor, o 7-de-hidrocolesterol ou provitamina D, presente naturalmente na pele, que age como precursor de metabólitos ativos que influem na formação e na conservação da função de barreira, na ativação de peptídeos antimicrobianos, nas atividades fotoprotetoras e na proteção contra o envelhecimento.
The skin innately possesses metabolic pathways and receptors to protect against external assaults. Here, one protective component, 7-dehydrocholesterol or provitamin D, naturally present in the skin, is described, which serves as the precursor for active metabolites that influence the formation and maintenance of barrier function, the activation of antimicrobial peptides, photoprotective activities, and protection against senescence.

Embora já seja conhecida desde 1922, a vitamina D3 ou colicalciferol só foi identificada mais recentemente, no final da década de 60, como precursora do esteroide ativo 1,25-di-hidroxivitamina D3 ou calcitriol. Esse esteroide ativo era conhecido por seu envolvimento no metabolismo do cálcio, ou seja, por seus efeitos sobre a mineralização óssea. Por isso, as crianças passaram a ingerir óleo de peixe, que possui altas concentrações da vitamina D3. A suplementação alimentar com vitamina D é comum hoje em dia, e beneficia toda a população.
Inicialmente, supunha-se que o metabolismo da vitamina D3 para calcitriol ocorria apenas no fígado e nos rins. Porém, mais recentemente, constatou-se que o receptor de vitamina D (VDR) para 1,25-di hidroxivitamina D3 está presente em mais de 30 tecidos ou órgãos, inclusive na pele, o que gerou novos estudos e relatórios sobre as atividades dos metabólitos da vitamina D e seu papel regulador de importantes funções celulares da pele.
